By co-hosting a few episodes of the Univision talk show Control (1994–1955) with former Entertainment Tonight anchor Carlos Ponce, De Pablo made her debut in the entertainment industry at the age of 15.
After graduating, she moved to New York City to pursue an acting career while supporting herself by working as a waitress. In the year 2000, she made appearances on The $treet on Fox and The Education of Max Bickford on CBS. She was cast in commercials, the All My Children television series, and the New York Public Theater. De Pablo is most known for her role as Israeli Mossad spy turned NCIS agent Ziva David in the police procedural drama NCIS.

After departing NCIS in 2013, she made a brief comeback in a regular role between 2019 and 2020. She has appeared in a number of films, however nothing particularly noteworthy. She and former NCIS castmate Michael Weatherly were named executive producers of the upcoming CBS drama MIA in 2018.
